Constructs for Transgenic Model Generation
Recombineering (Red/ET recombination) has become the method of choice to engineer large replicons, such as large targeting constructs or bacterial artificial chromosomes (BACs). Since recombineering can be performed at any position in DNA, the technology has opened up an unlimited number of possibilities for complex genetic modifications.
Gen-H’s portfolio encompasses the assembly of all types of constructs for animal models: targeting constructs, BAC transgenes, lentiviral-vectors, etc. We offer custom models containing point mutations, tags, reporter genes, gene knock-in (KI), knock-out (KO), and conditional constructs (cKI, cKO), as well has more complex designs, such as mini-genes and FLEX constructs.
Backed by 20 years of experience working with large, repetitive and challenging sequences, Gen‑H has specialized in developing large BAC-based targeting constructs, routinely assembling custom, complex constructs for humanizations of up to 240 kb.
Our scientists provide guidance in construct design and screening strategies for genotyping. Clear communication over the course of the project and full-coverage, nanopore-based sequencing of all completed targeting constructs ensures successful project completion.
OzBIG: Full-package mouse models with big genomic replacements
Through a successful long-standing partnership with Ozgene Pty Ltd., we are pleased to offer full-package mouse model development.
This partnership leverages the expertise of Gen-H in the assembly of large and complex constructs with the expertise of Ozgene (https://www.ozgene.com/) in mouse model development.
OzBIG is an advancement in gene targeting that combines the use of large targeting constructs with Ozgene’s enhanced gene targeting platform, incorporating its proprietary goGermline™ (https://www.ozgene.com/services/gogermline-knockout-and-knock-in-mice/) technology to significantly improve efficiency over conventional approaches. Together with a screening platform developed at Ozgene, this enables the reliable generation of ES cell clones carrying very large genomic modifications (20–240 kb). This process is performed without the use of exogenous recombinases like CRISPR.
OzBIG enables the generation of sophisticated mouse models with complete genomic loci up to 240 kb replaced by the human counterpart. These models are valuable tools for research in basic science, human disease, and drug discovery and development.
As of January 2026, more than 50 projects have been successfully completed (germline transmission confirmed).
